Stimulation of cells can produce calcium signals by initiating entry of Ca2+ ions across the plasma membrane or releasing them from intracellular stores. Activation of G-protein-coupled receptors (GPCRs), for example, leads to generation of the second messenger inositol 1,4,5-trisphosphate (IP3), which releases calcium from the endoplasmic reticulum. Alternatively, neurotransmitter stimulation of neurons can cause calcium to enter cells through voltage gated calcium channels in the plasma membrane.
Potential sites for pharmacological manipulation of calcium signalling therefore include: voltage gated and ligand gated ion channels, the ryanodine and IP3 receptor, Na+/Ca2+ exchangers in the plasma and mitochondrial membranes, and Ca2+ATPase in the plasma membrane.
