Name TMB ELISA Substrate (standard)
Biological description

Standard sensitivity HRP substrate for use in colourmetric ELISAs. Upon reaction with HRP the TMB ELISA Substrate (high sensitivity) develops a blue color with absorbance at 650nm. Upon stopping the reaction with 0.16M sulphuric acid the color changes to yellow which can be read at 450nm.

Protocol

  1. Add 100 µL of TMB ELISA Substrate (standard sensitivity) to each ELISA well,
  2. Incubate at room temperature for 15-30 minutes or until a visible color change has occured. The absorbance can be measured at this point if required at 650nm,
  3. Add 100µL of 0.16M sulphuric acid to each well to stop the reaction,
  4. Measure absorbance at 450nm on a plate reader.
